Transmission EM of cryo-immobilized samples * (TEM cryo samples)
In transmission electron microscopy (TEM) the accelerated beam of electrons is transmitted through the sample and collected to form an image. To achieve this the specimen is usually sectioned into ultrathin sections less than 100 nm thick. In this method, the samples are cryo-immobilising by means of high-pressure freezing to fix them in their "close to native" state. Specimens then go through a process of freeze substitution where they will be stained and eventually embedded into a resin for thin sectioning and imaging in the TEM.
